Laminin-521 facilitates self-renewal of both pluripotent human ES and iPS cells in a chemically defined, feeder-free and animal-protein free cell culture system. Importantly, Laminin-521 allows the survival and expansion of pluripotent human cells after plating from single cell suspension.
Laminin-521 (also known as laminin-11) is normally expressed and secreted by human pluripotent ES cells and can, in addition, also be found in the kidneys, neuromuscular junctions, lungs and placenta.

Single-cell passagingLaminin-521 facilitates survival and maintains pluripotency of hES and iPS cells after plating from single cell suspension. Pluripotent cells can be passaged by simple trypsinization into and re-plated as a single-cell suspension, with stable pluripotency marker expression for months. Laminin-521 supports long-term self-renewal at a stable rate higher than cells grown on e.g. Matrigel. More efficient and cost effective since, since you need less coating material per cell division and much less invested labor time. Due to the simplicity of the passaging procedure the experimental variance is reduced and the process can be automated for high-throughput cell culture and scientific results. |
BiorelevanceNo cell detrimental rho-kinase (ROCK) inhibitor treatment is needed to increase cell survival after single-cell enzymatic dissociation (Watanabe et al., 2007). Human pluripotent ES cells normally express Laminin-521 (Rodin et al., 2010; Miyazaki et al., 2008). Laminin-521 can increase the survival of dissociated pluripotent stem cells by conveying the highest dose of signal via a6b1 integrin activating the PI3K/Akt pathway. Human ES and iPS cells plated on Laminin-521 grow as a monolayer, which makes the culture homogenous since cells are equally exposed to the matrix and culture medium. |
